Hog kidney gluconokinase.

چکیده
Previous studies (1, 2) have indicated that gluconate is extensively metabolized in the mammal without, prior conversion to glucose. The int, erpretation of experiments performed with labeled gluconate has rested upon the assumption (3) that gluconate is converted to 6-phosphogluconate in t,he course of its metabolism. The enzyme, gluconokinase, which catalyzes this reaction has been partially purified from Escherichia coli (4) and yeast (5), but has not previously been demonstrat,ed in mammalian systems. In the present study, the phosphorylat, ion of g1uconat.e according t,o Equation 1 has been shown to occur in extracts of mammalian tissues and the enzyme, gluconokinase, has been partially purified from hog kidney.

Studies with Rat Liver Glucose Dehydrogenase.
Glucose dehydrogenasel catalyzes the direct oxidation of glucose to &gluconolactone (2) utilizing as a coenzyme either nicotinamide adenine dinucleotide (NAD) or nicotinamide adenine dinucleotide phosphate (NADP) (3-5). Harrison (6,7) first reported its presence in cat, dog, sheep, and ox livers in 1931 and later achieved a crude purification of the beef liver glucose dehydrogenase (8). Improve...

The study was carried out to detect and describe the outbreaks of Hog Cholera (HC) or Classical Swine Fever (CSF) in different parts of NorthEastern region of India by histopathological techniques in post mortem samples. Histopathological alterations in different lymphoid and non-lymphoid organs provides clue to confirm hog cholera disease. Pigs died of suspected swine fever (hog cholera) virus...

Hog kidney Acylase I, an enzyme which hydrolyzes N-acetylL-amino acids, has been purified approximately 40-fold by Greenstein (1). During the course of the purification of hog kidney dialkylfluorophosphatase, it was consistently found that the protein fractions containing DFPasel activity also possessed very high Acylase I activity. Inasmuch as the DFPase and Acylase I activities were found to ...
متن کاملBiochemical Characterization of Human Gluconokinase and the Proposed Metabolic Impact of Gluconic Acid as Determined by Constraint Based Metabolic Network Analysis
The metabolism of gluconate is well characterized in prokaryotes where it is known to be degraded following phosphorylation by gluconokinase. Less is known of gluconate metabolism in humans. Human gluconokinase activity was recently identified proposing questions about the metabolic role of gluconate in humans. Here we report the recombinant expression, purification and biochemical characteriza...
متن کاملRegulatory mutations affecting the gluconate system in Escherichia coli.
A spontaneously arising regulatory mutant of the gluconate system in Escherichia coli was isolated. This mutant became constitutive, probably in one step, for gluconate high-affinity transport, gluconokinase, and gluconate-6-P dehydrase. The mutation involved (gntR18) is cotransducible with asd. Pseudorevertants, derived from a mutant (M2) that shows a long lag for growth on gluconate mineral m...
